Traffic on the stretch had to be diverted for over two hours on Thursday.
Madurai Rural Police said that the bus, which was coming from Tuticorin to Madurai, collided against the lorry coming in the opposite direction.
In the impact, the bus was heavily damaged and it overturned on the middle of the road.
One person identified as Bharath (35) of Odisha, who was travelling in the bus died, while seven other passengers suffered multiple injuries.
They were all rushed to the Government Rajaji Hospital for treatment.
Due to the accident, vehicular movement on the stretch was blocked for over two hours, the police said and added that the traffic was diverted through the city at two of the Ring Road intersections.
Due to some delay in communication, the rescue operation could not be carried out swiftly.
Preliminary inquiries with the passengers suggested that over speed, rash and negligent driving was a cause.
Skeleton strength of police at the spot posed a tough time in regulating the vehicles. Many trucks and heavy vehicles had lined up on both sides, a senior official said.
